Eluent conditions for Asahipak NH2P-50:
HILIC mode
(1) Water, acetonitrile, and ethanol, either alone or in mixtures of any ratio, may be used as eluent. With aqueous ethanol solutions, the flow rate should be lower than normal, because of their relatively high viscosity.
(2) Various types of buffers soluble in acetonitrile or ethanol may be used together with the above components, as long as no observable precipitation occurs in the eluent.
(3) Eluents of pH 2 to 13 can be used.
Anion-exchange mode
(1) Buffer solutions such as phosphate, acetate, and tris, with or without NaCl, KCl, or Na2SO4, may be used.
(2) Eluents of pH 2.0 to 13.0 can be used.
The packing material for Asahipak NH2P-50 columns has amino groups which are bonded to weak anion exchange resins. The amino groups exist in the equilibrium (free base ratio) of free type and acid type.
The state of the equilibrium depends on pH and ionic composition of the eluent. The elution times became shorter and peak shapes became sharper as free base ratio increased with higher pH.
